Monoclonal antibody CA 15-3 reacts with the DF3-antigen, a high molecular weight mucin-like glycoprotein. The DF3 antigen is present as a 300 kD species on the apical borders of secretory mammary epithelial cells. The antigen has also been reported to circulate as a heterogeneous antigenic species with molecular weights ranging from 300 to 450 kD. The binding site for monoclonal antibody CA 15-3 involves a glycosidic linkage between carbohydrate and protein moites on the DF3 antigen.
